Autophagy by podocytes in renal biopsy specimens.

چکیده
Autophagy, a process of bulk protein degradation and organelle turnover, is induced under starvation conditions in yeast and plays an important role in developmental processes and human diseases. Autophagy is a nonapoptotic form of programmed cell death. Disturbances in autophagy and programmed cell death may lead to cancer and several degenerative diseases in humans. During the routine ultrastructual examination of renal biopsy, specimens we found two types of autophagy in podocytes. The presence of autophagy in patients with renal disease might represent a change in physiological conditions to prevent podocyte damage. Of the renal diseases with autophagy in podocytes, type I autophagy was found in 10% of cases and type II autophagy in the remaining 90% of cases. The presence of type I autophagy was not clearly correlated with age, sex, or pathological diagnosis. However, many cases of type I autophagy were associated with a long history of treatments for renal disease. Thus, the appearance of type I autophagy might suggest a poor prognosis. Protein and lipid clearance were greater in type II autophagy than in type I autophagy. However, whether type I autophagy evolves into type II autophagy or whether a mutation in the autophagy gene occurs is unclear.

Correlation of autophagy type in podocytes with histopathological diagnosis of IgA nephropathy.
OBJECTIVE IgA nephropathy (IgA-N) frequently leads to progressive renal failure, thus estimation of the degree of progression is important for patient management. Autophagy is a mechanism that facilitates clearance of waste products to preserve renal function. The aim of this study was to assess autophagy in podocytes in children with progressive IgA-N at initial diagnosis by electron microscop...
متن کاملPathogenic old world hantaviruses infect renal glomerular and tubular cells and induce disassembling of cell-to-cell contacts.
Viral hemorrhagic fevers are characterized by enhanced permeability. One of the most affected target organs of hantavirus-induced hemorrhagic fever with renal syndrome is the kidney, and an infection often results in acute renal failure. To study the underlying cellular effects leading to kidney dysfunction, we infected human renal cell types in vitro that are critical for the barrier functions...
متن کاملRedistribution of connexin43 expression in glomerular podocytes predicts poor renal prognosis in patients with type 2 diabetes and overt nephropathy.
BACKGROUND Significance of podocyte injury in the progression of diabetic nephropathy is not well-understood. In this study, we examined whether alteration of gap junction protein connexin43 (Cx43) expression in podocytes is associated with the progression of overt diabetic nephropathy. METHODS We recruited 29 type 2 diabetic patients with overt nephropathy who underwent renal biopsy. Nephrec...
متن کاملApelin involved in progression of diabetic nephropathy by inhibiting autophagy in podocytes
Podocyte autophagy dysfunction has been reported to be responsible for the progression of diabetic nephropathy (DN), however, the factors contributed to autophagy dysfunction in type 2 diabetes are not fully understood. Among promoting factors in DN, an adipokine, apelin, had been showed to trigger podocyte dysfunction. Therefore, it is hypothesized that apelin, which is increased in plasma in ...
متن کاملSoluble epoxide hydrolase in podocytes is a significant contributor to renal function under hyperglycemia.
BACKGROUND Diabetic nephropathy (DN) is the leading cause of renal failure, and podocyte dysfunction contributes to the pathogenesis of DN. Soluble epoxide hydrolase (sEH, encoded by Ephx2) is a conserved cytosolic enzyme whose inhibition has beneficial effects on renal function. The aim of this study is to investigate the contribution of sEH in podocytes to hyperglycemia-induced renal injury. ...
